RESUMO

Objective:To develop a rapid, simple and cost-effective quantitative TaqMan RT-PCR (RT-qPCR) that could be used as an alternative to sequencing for the detection of Omicron variants and to evaluate its performance. Methods:Primers and TaqMan probes targeting the conserved domains of SARS-CoV-2 ORF1ab and the high-frequency mutation sites in the S gene of Omicron variants were designed. Then a RT-qPCR for the detection of Omicron variants was established. The consistency of the method was verified using samples identified by whole-genome sequencing. The specificity and sensitivity of the method were also evaluated. Results:The established RT-qPCR could distinguish Omicron variants from early epidemic A strains and Alpha and Delta variants of SARS-CoV-2, and the results were consistent with those of whole-genome sequencing with a coincidence rate of 100% (28/28). There was no cross-reactivity with other six respiratory viruses or coxsackievirus group A16. For RNA standards, this method showed good linearity in the range of 10 9-10 3 copies/μl with a correlation coefficient ( R2) greater than 0.99 and detection sensitivity of 10 3 copies/μl. Conclusions:The RT-qPCR designed in this study for Omicron variant detection had good sensitivity and specificity and could be easily performed in laboratories, which would greatly facilitate the monitoring of Omicron variants.

RESUMO

Objective:To evaluate the effectiveness of the course of Laboratory Medicine and Clinical Sciences in the Laboratory Medicine Faculty of Sun Yat-sen University. Methods:Twenty-four undergraduate students in Batch 2016 of Medical Laboratory Faculty were divided into small groups (4-6 students per group). They learned each case in groups before class. In the first session of each class, the case-based study (CBS) tutor would randomly assign case-related questions to the students. Students were required to present their answers in class. The CBS tutor would guide the students to discuss the case further. In the second session, a lecture associated with the case would be given by a special subject lecturer. After the course, students, tutors and lecturers were given questionnaires and were randomly interviewed to comprehensively understand the course's effectiveness. SPSS 19.0 was used for statistics. Results:Most case discussion tutors agreed that they could guide students to discuss clinical cases well in class and give comments according to students' presentations (93.75%, 15/16). Most of the lecturers agreed that they could well guide students to think about the relationship between laboratory and clinical diagnosis and treatment in class (91.67%, 11/12). Both teachers and students had very positive evaluations of the learning mode, learning content, inspiration to students, and teachers' ability of this course. All the teachers and students agreed that the learning mode of combining CBS with special subject lecture was more helpful for the students to systematically learn medical knowledge compared with a CBS session alone or a lecture alone. Conclusion:The course, Laboratory Medicine and Clinical Sciences, which combines the CBS with the traditional lecture mode, integrates the advantages of the two learning modes. It not only stimulates students' enthusiasm for active learning, deepens clinical knowledge memory, and builds a clinical thinking model, but also enriches the teaching modes of medical laboratory education.

RESUMO

Background@#Excessive delivery of free fatty acids (FFAs) to the liver promotes steatosis and insulin resistance (IR), with IR defined as reduced glucose uptake, glycogen synthesis and anti-lipolysis stimulated by normal insulin levels. Whether the associations between FFAs and diabetes development differ between patients with and without nonalcoholic fatty liver disease (NAFLD) remains unclear. @*Methods@#Consecutive subjects (2,220 NAFLD subjects and 1,790 non-NAFLD subjects according to ultrasound imaging) were enrolled from the First Affiliated Hospital of Sun Yat-sen University between 2009 and 2019. The homeostasis model assessment of insulin resistance (HOMA-IR) was calculated. @*Results@#There was an approximate J-shaped relationship between FFA levels and HOMA-IR in the NAFLD group. Higher FFA concentration quartiles were associated with higher risks of IR (odds ratio [OR], 9.24; 95% confidence interval [CI], 6.43 to 13.36), prediabetes (OR, 10.48; 95% CI, 5.66 to 19.39), and type 2 diabetes mellitus (T2DM; OR, 19.43; 95% CI, 12.75 to 29.81) in the NAFLD group but not in the non-NAFLD group. The cut-off points for the FFA levels increased in a stepwise manner in discriminating IR, prediabetes and T2DM (573, 697, and 715 μmol/L) in the NAFLD group but not in non-NAFLD individuals. @*Conclusion@#A distinct dose-dependent relationship of FFA levels was found with IR, prediabetes and T2DM in NAFLD patients. Screening serum FFA levels in NAFLD patients would be valuable in preventing diabetes development.

RESUMO

ObjectiveTo assess the value of quantitative tissue velocity imaging (QTVI) in monitoring the establishment of levothyroxine-induced cardiomyopathy models on rabbits.MethodsThirty New Zealand purebred rabbits were divided into experimental group (n=20) and control group (n= 10).Hyperthyroidism models were established by peritoneal injection of levothyroxine (L-Thy) in experimental group (45 μg/ [kg · d]).The same dose of saline was given to rabbits in control group for 4 weeks.QTVI and conventional echocardiographic parameters were obtained at the beginning day and 1,2,4 weeks in two groups.According to the changes of ultrasound parameters, the rabbits of experimental group were divided into two subgroups:Concentric hypertrophy (CH) subgroup and eccentric hypertrophy (EH) subgroup.ResultsAt the first week, left ventricular ejection fraction (LVEF),fractional shortening (LVFS),mitral inflow peak velocity (E),average mitral annularsystolic peak velocity (Vs) and acceleration (a) increased obviously than before injection in experimental group (P<0.01).At the second week, posterior wall thickness at end diastole (PWTd),interventricular septal thickness at end diastole (IVSTd) and left ventricular mass index (LVMI) increased obviously than before injection in experimental group and control group (P<0.05),and the average mitral annular early diastolic velocity (Ve) was significantly lower (P<0.01).Compared with control group, the average Ve was significantly lower in CH subgroup (P<0.01),while LVEF,LVFS,average Vs,a and average Ve were significantly lower in EH subgroup (P<0.01).ConclusionQTVI is valuable in the assessment of establishing levothyroxine-induced cardiomyopathy models on rabbits.

RESUMO

Objective: To probe the effects of gross polysaccharide of Radix Rehmanniae Preparata(GPRRP) on the morphology changes of immune organ of blood deficiency model mice. Methods: Using the blood deficiency mice models induced by releasing blood and injection of Cyclophosphamide as the experimental objects, the model animals were divided into 5 groups, that was the large, middle and small dose of GPRRP groups,Danggui Buxue Oral Liquid group, model group,at the same time, giving a blank control group. It was given successively relating drugs to the animals of several groups for 10 days.At the end of the experiment, the hemogram was measured and the thymus and spleen were taken, and making pathological section, then these tissues morphology changes were observed under microscope. Results: GPRRP can obviously resist the atrophy of thymus and spleen induced by the model establishing, can obviously increase the depth of thymus cortex and the number of cortex cells, can enlarge the size of spleen nodules and obviously increase the number of lymphocyte. Conclusion: GPRRP can resist the atrophy of thymus and spleen in blood deficiency model mice.
